Caracterización fenotípica y genotípica de germoplasma de Panicum coloratum L var. coloratum para la tolerancia a la salinidad

Los cultivares de Panicum coloratum son poblaciones estabilizadas que presentan un cierto grado de variabilidad intracultivar. Plantas individuales provenientes de pasturas establecidas en suelos salinos, afectados además por condiciones de extrema sequía, pueden ser fuente de germoplasma con tolerancia a estreses múltiples. El objetivo del presente trabajo fue identificar la variabilidad para tolerancia a la salinidad en 16 individuos de P. coloratum var coloratum provenientes de una pastura establecida en la Pampa Deprimida, provincia de Buenos Aires, que sobrevivieron a sequía prolongada y fuertes heladas. Los materiales se multiplicaron vegetativamente en invernadero y se realizaron ensayos en condiciones controladas de luz y temperatura. Esquejes homogéneos enraizados, se cultivaron sobre sustrato inerte de arena y perlita con suministro de solución nutritiva. Se administraron tres niveles de salinidad (0, 200 y 400 mM NaCl) y se evaluó crecimiento vegetativo durante la etapa de crecimiento vegetativo inicial. Las variables analizadas fueron tasa de aparición de hojas, tasa de macollaje, peso seco aéreo, número de hojas y macollos finales, además se evaluó concentración foliar de Na+ y K+. La tolerancia a la salinidad fue evaluada por la comparación del crecimiento en presencia salinidad respecto al de ausencia de salinidad, usando una o más variables de crecimiento. El análisis de crecimiento indico variabilidad entre clones en tolerancia a la salinidad y crecimiento La jerarquización de los materiales de acuerdo a la tolerancia fue calculada teniendo en cuenta una o varias variables de crecimiento obteniendo resultados similares. También se identificó variabilidad genética entre los clones mediante marcadores moleculares ISSR. La variabilidad identificada podría ser usada en programas de mejoramiento genético vegetal para obtener poblaciones con mayor tolerancia a la salinidad en esta especie.

nity, severe droughts and frost in a plot established in the
Depressed Pampa
, in the
province of Buenos Aires, Argentina. Plants were vegetatively cloned and trials were
conducted under controlled greenhouse conditions, using homogeneous cuttings grown on
an inert substrate of sand and perlite and irrigated with nutrient solution.
Plants were
treated with three salinity concentrations (0, 200 and 400 mM NaCl) and evaluated for
responses at the initial stages of vegetative growth. Shoot dry weight, number of
leaves
and tillers, rate of emergence of leaves and tillers and leaf Na
+
and K
+
concentrations were
evaluated.
Salt tolerance was assessed by comparing growth under saline conditions to
growth in the absence of salinity, using one or more growth variables
.
The analysis of
growth data indicated variability among clones both in terms of growth and salt tolerance.
Salt tolerance ranking using one or more variables shed similar results. Genetic variability
among clones was assessed by inter simple sequence rep
eat analysis (ISSR).
The
variability detected in this study suggests the collected plants could be adequately used in
a breeding scheme to increase salt tolerance in this species.
